Principle:- Peptone, serves the simplest source of the nitrogen, carbon and other growth factors such vitamins. Di-sodium phosphate acts as buffering agent, and sodium chloride maintains the osmotic balance. Buffered Peptone water is designed to help recovery of the sub-healthy damaged Salmonellae from processed food samples before transferring to selective medium. Buffered Peptone water has showed higher recovery of Salmonellae than that of the Lactose broth. Directions:- Suspend 20.0 gram in 1000 ml distilled water adjust the pH if required. Boil to dissolve the Buffered Peptone Water medium completely. Sterilize by autoclaving at 15 lbs pressure (121 °C) for 15 min, cool.
